Can the output shaft be up and vertical so as to adapt to the rear of a drill press?
A: There optional breather vent ports to allow that orientation.

Can the faster turning shaft be point up in a vertical orientation and the slower moving shaft point in a horizontal direction. The application is to hand crank the slower shaft and have the faster shaft turn a winding spindle for very light weight, fairly delicate material. Thanks,
A: These are worm gear drive boxes and would not be easily back-driven in that fashion by hand. With the 5:1 reduction, it can be done but the continual back driving may damage the gear and the worm in fairly short order and greatly reduce the lifespan of the box.

Can unit be used in reversed to amplify speed? Example turn 100rpm into 500 rpm?
A: No, it could not.

will it work to put two gear reduction units in line to reduce the speed and increase torque
A: Yes, as long as the output torque on the last unit does not exceed the mechanical ratings of the box itself.
